Streptomycin stock solution (20 mg ml-1)(50 ml)
1. Weigh out 1 g streptomycin sulfate and dissolve in
45 ml of distilled water.
2. Sterilise by filtration.
<STORAGE> Aliquots of appropriate volume
can be stored at -20 C.
<USAGE> For culture plates, allow media to cool to 55 C before adding streptomycin to a final concentration
of 25 ug/ml.
Tetracycline stock solution (12.5 mg ml-1)(80 ml)
1. Weigh out 1 g tetracycline and
dissolve in 75 ml of 1:1 vol/vol distilled water:ethanol.
2. Make up volume to 80 ml with 1:1 vol/vol
distilled water:ethanol.
3. Sterilise by filtration.
<STORAGE> Aliquots of appropriate volume should
be wrapped in aluminium foil and stored at -20 C.
<USAGE> For culture plates, allow media to cool to 55 C before adding streptomycin
to a final concentration of 12.5-15 ug/ml. Store plates in the dark at 4 C. Magnesium ions are antagonists of tetracycline, therefore
avoid using media, such as LB that contain magnesium salts.
1. Weigh out
25 g Luria broth powder and dissolve in 750 ml of distilled water.
2. Add 800 ml distilled water to
dissolve.
3. Adjust to pH 7.0 with either HCl or NaOH as appropriate.
4.
Make up volume to 1 liter with distilled water.
5. Sterilize by autoclaving for 15 min at 121 C.
<STORAGE>
Store at 4 C.
1. Measure 90 ml distilled water.
2.
Add 6.8 g Na2HPO4.7H2O, 3 g KH2PO4, 0.5 g NaCl and 1 g NH4Cl sequentially, allowing each salt to dissolve before adding the next.
3. Make
up volume to 100 ml with distilled water.
4. Sterilize by autoclaving for 15 min at 121 C.
<STORAGE>
Store at room temperature.
1. Weigh out 1 g NH4Cl and 3 g glucose.
2. Add 100 ml 10X M9 Salts.
3. Add 1 ml 1 M MgSO4 and 10 ml Gibco MEM vitamin solution.
4. Make up volume to 1 liter with distilled water.
5. Check that the pH
is 7.0-7.4.
6. Filter sterilize through a 0.22 micron filter.
7. Add 1
ml sterile 100 mM CaCl2 solution.
<STORAGE> Store at room temperature.
