Immunoaffinity Fractionation Protocols (from GenWay)

 

Separation or fractionation of complex protein mixtures that have an overwhelming dynamic range of concentration, such as plasma or serum, is an outstanding challenge for proteomic analysis. Sample preparation to remove highly-abundant proteins is an essential step for enhancing detection resolution by two-dimensional gel electrophoresis (2DE), liquid chromatography (LC) and/or mass spectrometry (MS). Various tools and methods for protein separation using affinity capture reagents are capable of specific binding and target removal. Among these reagents, immunoaffinity mediated separation of protein mixtures is the classical, natural, effective, and most reliable approach. Antigen affinity-purified polyclonal avian (chicken) IgY antibodies, compared to the more conventionally-used IgG antibodies, have unique and advantageous features that enable specific protein separation and sample preparation for low-abundant protein detection and biomarker discovery. To best meet the needs of proteomic analysis by 2DE and MS, it is desirable that sample preparation tools for protein separation or fractionation have the following features: High specificity and accuracy
Low cross-reactivity to non-target proteins
High avidity (strong binding power)
Bound proteins can be eluted and analyzed
High capacity (to process large volume per load)
Multi-species applicability
Convenient use (simple and easy operation)
Can be automated or streamlined
High reproducibility (with acceptable CV)
Good reusability (fidelity in regeneration)
Minimal disruption to natural condition of sample
